Abstract Serotonin-dopamine antagonists (SDAs) are novel antipsychotics which have different pharmacological properties from conventional antipsychotics. SDAs possess potent 5-HT2A receptor and weak D2 receptor antagonism, whereas conventional antipsychotics such as haloperidol blocks D2 receptors with higher affinity than 5-HT2A receptors. From their distinct receptor profiles, SDAs effectively reduced negative symptoms and produced less extrapyramidal side effect than conventional antipsychotics, however they are equally potent to conventional agents in treating positive symptoms. A number of SDAs drugs are being developed which will provide new modalities in treating schizophrenic patients and their clinical efficacy remains an issue to be verified.
